通过机制建模捕捉 CAR-T 细胞疗法的动态变化

摘要: Chimeric antigen receptor T (CAR T)-cell therapy has shown remarkable success in treating various leukemias and lymphomas. CARs are engineered to redirect T cells to specific antigens. Much is still unknown about the workings of CAR-T cells in the body, and there is not a standard monitoring process. Modeling can shed light on CAR T-cell PK/PD and inform future studies. We developed a mechanistic model to capture CAR T-cell PK and validated our model with PD results. Our model can reproduce PD and provide information on characteristics of CAR T-cells and B cells that drive system dynamics. Furthermore, sensitivity analysis reveals patient and drug qualities that could improve tumor killing and subsequent outcome.
